Describe and explain the different responses of the characters to the Inspector’s visit; and explore Priestley’s reasons for showing a difference between the older and younger generations  The characters respond to the Inspector’s news in very different ways. You get the impression that Arthur Birling doesn’t care about the situation that has occurred with Eva Smith.  Ã¢â‚¬ËœAnd I don’t see where I come into this.’ This suggests that he thinks that he cannot do wrong but we find out that he is the one that sacked Eva Smith for wanting a pay rise. This started the sequence of events that led to her eventual death. Arthur Birling is moved to anger. He implies that he is a friend of the Chief Constable Colonel Roberts and that the Inspector cannot talk to him in such a way. He considers the talk as ‘uncalled for and officious’. Arthur tries to belittle the Inspector and is very hostile towards him. He reacts like this because he doesn’t want to ruin his reputation and blames the Inspector for the interruption of the celebration of Sheila and Gerald’s engagement. Arthur’s social position makes him react as he does. He thinks that he is superior and only looks after himself. Birling is presented to us as a character that thinks he is right and won’t be tolerant Birling’s daughter Sheila on the other hand is co-operative with the Inspector. She notes that no one told him anything that he did not already know. She is moved to tears of shock and guilt: the Inspector is successful in making her see the consequences of her actions. When she realises that she is the person that got Eva Smith sacked she thinks that she is responsible for her death. Sheila is more supportive to the enquiry. This shows that she is understanding and shows that she cares about the whole Eva Smith situation, unlike her father Mr Birling. Here we can start to see the different attitude that the younger and older characters play. Arthur Birling keeps insisting that Sheila should not be brought into this matter. Mrs Birling says to Sheila ‘I think you ought to go to bed’. She is trying to control Sheila and treats her like a child. Mrs Birling thinks that she is superior ‘Girls of that class -‘it is as though she is looking down on the girls. Regardless of the fact that Gerald, Mr Birling and Mrs Birling try to protect Sheila; she is the one that can actually face the truth. When the Inspector says that Eva Smith was also known as Daisy Renton, Gerald tries to hide his reaction because he played a part in Daisy Renton’s life. He is distressed by his realisation in Eva Smith’s/Daisy Renton’s life and death. Sheila tells him to tell the Inspector, implying that the Inspector knows the truth already. Gerald asks Sheila to leave the room because he doesn’t want to expose any more unpleasantness. Gerald tries to justify that he is a hero when he says:  Ã¢â‚¬ËœI became at once the most important person in her life†¦Ã¢â‚¬â„¢Ã‚  whereas he is also anxious that he has a shameful link to Eva Smith’s death. Mrs Birling remains entirely untouched by the Inspector’s questioning.  Ã¢â‚¬ËœI did nothing that I’m ashamed of or that won’t bear investigation.’  She feels no shame because she refuses to see how Eva Smith’s death can have followed as a consequence of her actions. Mrs Birling believes that a working class girl deserves all that she gets. She reveals her assumptions about people ‘make sure that he’s compelled to confess in public his responsibility’. She thinks this is the right thing to do but at the end she doesn’t do the right thing.  When the inspector confronts Eric, he reacts similarly to his sister, realising that he has done wrong. Eric is a complicated character. At the start of the play he is really immature. He believes that women are just there to be sexually used. When he gets Eva pregnant he tries to do the decent thing by giving her money. He steals the money from Birling’s office.

The three poems Remember by Christina Rossetti, Plena Timoris by Thomas Hardy and Refugee Mother and Child by Chinua Achebe all portray death in different ways. Rossetti explores the acceptance of death, whereas, in Refugee Mother and Child we see the unwavering love the mother has for the child. In the poem Plena Timoris the persona recalls how love can be so extreme that could lead to despair and death. What I found intriguing about these poems was how love could be both a dangerous and positive force towards the subject of death. Love could drive a person to their grave but also love could be the will for a person to stay strong and never giving up. Christina Rossetti a devote Anglican to whom church and family were paramount. It is known that two marriage proposals which did not live up to her religious fervour therefore she turned them down. Because of her religion Rossetti appears to have been obsessed with her own pending death in this poem. Her belief was that death was the ultimate goal for everyone, and because of such she very much planned her own pending death. The structure of this poem was in a Pertrachan Italian sonnet form. In this form poems are meant to be very structured and are need of following a strict rhyme scheme. However there was a change in rhyme scheme between the octet and sestet. In the octet the phrase â€Å"Remember me† was mentioned three times, but a sudden change was then noticed, as in the sestet â€Å"Forget me† was repeated twice. This change is used to suggest the possibility of forgetting and being happy rather having instructions to remember. This could show even before her death she was still doubtful between secular love and materialistic love. Chinua Achebe is a proud Nigerian who observed the civil war in his country and horrific suffering of the innocent refugees. In Refugee Mother and Child he relates the sadness of death between a child and a mother by writing in free verse. The line length of the whole entire poem was three lines. In the first stanza there is only one sentence, and in the last stanza two sentences were present. These long sentence increases the intensity on the images and also the enjambment suggests the suffering of these innocent refugees. Plena Timoris is set within the conservative morality of Victorian England where women had a subordinate and dependent position in a male patriarchal world. Hardy uses different tones throughout the poem to evoke how he thinks about love. He sees love in a very cynical view and is making an effort in portraying love is not everlasting but tragic and indecisive. In the first stanza the use of alliteration in â€Å"the lovers looked over† emphasizes love was lyrical and a harmony of life by the â€Å"l† sound produced. But in the second stanza onwards there was a sudden transition and the change of tone was turned emotionless and insensitive. From the phrase, â€Å"There’s a woman in the canal below† there is a sense of bluntness and matter of fact to it. This sudden contrast of tones between the stanzas just enforces how Hardy thinks about love. The rhyme scheme of this poem does not change, the structure is: â€Å"abbab, cddcd†. It is in a strict and orderly fashioned. This rhyme scheme reflects how Hardy feels abut love and death. That love would have happy times but at the end it will just lead to despair. Also by having such a strict rhyme scheme it also suggest how Hardy views women opinion in love. As this structure is so old fashioned it provides a kind of security and how the future is being set. This may be very well how women in the male patriarchal world viewed love therefore he viewed love in such a cynical way. In the poem Remember, Rossetti portrays death as an eternal and a beautiful thing, instead of fearing death like the other two poems. The persona appears to have been planning and waiting for her own pending death. Rossetti uses strong imagery to reveal about how she feels about death. The opening two lines of this sonnet â€Å"Remember me when I am gone away, Gone far away into the silent land† gives an idea of how she hints that she is dying. The phrase â€Å"silent land† could be translated as a cemetery or individual grave. It can also be seen as a metaphor as the phrase also suggests calm after life. All these techniques used by Rossetti could be seen that she is using euphemism to make death sound less harsh, and instead it is a goal that should be achieved by everyone. Whereas in the poem Refugee Mother and Child, Achebe reflects the sadness of death by creating a tragic atmosphere. This poem does so by introducing shocking images and strong words. In this poem it is full of pathos; it is a very emotional poem. The alliteration used in: â€Å"struggling in labored steps behind blown empty bellies† evokes a sense of harshness, as the â€Å"b† sound which is constantly repeated makes a very plosive sound. The juxtaposition used in â€Å"the rust – colored hair left on his skull† emphasizes how horrific death is towards the child. Not only is this phrase very descriptive but it also seems to describe the layers of death. It is very photo journalistic which makes the atmosphere very tragic and grave. In Plena Timoris death is conveyed in the strong imagery created by Hardy. The negative imagery in â€Å"And her arm dropt† symbolizes how because the woman in the poem sees love could lead to death it is in fear of it, therefore there is a loss of affection towards the man. The alliteration in â€Å"dim dreads† and â€Å"slowly sees† portrays a sense of insecurity coming from the woman. As she slowly sees how extreme love could lead the despair and death. Lastly, the phrase â€Å"till an arm upbore† in stanza two is a horrifying image. The word upbore gives a sense of death and the whole phrase conveys a sense of fear towards the subject death as it is being described as horror and terror. In Remember the poem creates a huge impact on readers. It is a poem which is very memorable. This is because Rossetti describes death in a very different way than most poets therefore standing out from the other poems. The repetition in this poem emphasizes the point of her point of view and the confusion between her religion and materialistic love. The phrases â€Å"Remember me† and â€Å"Forget me† was repeated a number of times and because of such it plays a great role in the poem. As it gives an impression on how Rossetti is judging between her church and her religious fervor. The strong descriptive imagery in the poem Refugee Mother and Child and the choices of word Achebe chooses makes this poem have a very strong impact on the reader. The beginning of the poem where allusion is used in: â€Å"No Madonna and Child could touch† by referring to a famous painting of Jesus and Mother Mary, emphasizes the point of love between the bonds of the mother and child very strongly. Also the strong imagery in â€Å"her eyes the ghost of a mother’s pride† gives an impression of how the mother is hanging onto the child’s life and as the child dies something inside the mother is dying too. These strong images are used often in Achebe’s poem, and because of such his poem have a strong impact on readers and this poem is very memorable. In Hardy’s poem it was very effective as the negative poetic techniques he uses describes about love in a very vivid and strong, also Plena Timoris gives an impact to readers because it shows very clearly the differences between love and death. The juxtaposition in† till hopeless despair began† gives a contrast between hope. By this phrase love is being described very negatively, and because of this the poem is very vivid in the readers mind. Also in the phrase â€Å"the girl’s heart shuddered; it seemed as to free her†. The semi colon used in this phrase is an intended pause to emphasize the sense of the women’s heart actually shuddering. Plena Timoris creates a strong impact on readers as the descriptive words he chooses allows the reader to imagine clearly what is happening. Personally I liked Refugee Mother and Child the most because Achebe boldly states that the human spirit and dignity of the Nigerian mother and the love she has for the child is greater than anything comparable in the Christian world. Though Hardy’s and Rossetti’s poem was not unforgettable pieces of work, I think Achebe’s is the better one because his comparisons stood out more than the ones Hardy had. Also the imagery he had and alliteration was stronger than Rossetti’s. By reading Achebe’s poem I felt and sympathized for the mother and child, and I thought the poets point was put forward very clearly on love and how strong it can be.

West Timor was a part of Indonesia and East Timor was a Portuguese controlled colony. East Timor was taken under the control of the Portuguese in the 1600s. They were in control until 1975. During 1942 the Japanese invaded East Timor whilst fighting the Australian troops. Around 60,000 East Timorese were killed for protecting the Australian troops. The Japanese were in control of East Timor until 1945. In 1975 on the 28th of November, the Fretilin declared East Timor as independent. On the 5th of December that year the Indonesian forces invaded East Timor; claiming it as a province. The U. N. did not officially recognise the move by the Indonesians. The East Timorese tried to resist the Indonesian invasion, all attempts failed. While the invasion was happening 200,000 East Timorese died. The political climate in 1975 The fear of communism had spread throughout Asia at the Vietnam War. Many countries were afraid of it and the Indonesians accused the East Timorese of trying to become communist. The Indonesian military used this as a scare tactic for the rest of the world. They claimed that they were only trying to keep East Timor under control. But really they just didn’t want them to become independent. The Australian government knew of the invasion but did nothing to help the East Timorese people. The Hawke government didn’t want to risk Indonesia making us enemies. At the time, the Indonesian government had military, political and economic support from countries such as the UK, USA and Australia. It is to be believed that these countries did not aid the East Timorese from the invasion for various reasons, such as the Timor gap, trading and cheap labour interests. The impact of the invasion in 1975 and the future repercussions The invasion of the Indonesians left peoples homes destroyed, their family’s dead and their lives uprooted. Many refugees left East Timor in search of a peaceful new home. In August 1999, the 99% of the people of East Timor voted in a U. N referendum. Four days after the referendum the tally showed that 78% of the East Timorese voted for independence. A militia leader said it bluntly, Peace? Why would we want peace? If the vote is for independence well just kill—kill everybody. Within one week the Indonesian military-backed militia started a terror campaign. Women, children, but mostly men, boys, the educated, nuns, and priests were murdered. The Indonesian government was trying to make it so that East Timor didn’t have any educated people left. The capital, Dili, was set alight as people fled. Homes, churches and even the United Nations compound were attacked. Many refugees left for other Indonesian islands. The only safe places for the East Timorese were the four cantonments in the mountains that were held by the East Timorese armed resistance. At some points in time there were said to be 200,000 to 300,000 refugees with up to 600,000 people displaced. The role of significant individuals Jose Ramos-Horta: Jose Ramos-Horta was born and raised in East Timor. In 1969 he worked as a journalist and in 1974 he was exiled to Mozambique after his attempts to make East Timor independent anger the colonial administration. When Jose Ramos-Horta returned to East Timor he joined the Fretilin in 1974. In October 1975, Jose Ramos-Horta took the group of five journalists to the town Balibo so they can film the Indonesian attack. He left the town only hours before the five men were killed. On the 28th of November that year Jose Ramos-Horta was appointed as the Fretilin’s minister of communications and external affairs. Ramos-Horta had left just days before Indonesia invaded on the 7th of December. He was in New York representing East Timor to the UN Security Council. He successfully made a pass on the resolution demanding that Indonesia withdraw. But Indonesia ignored the UN. From 1977 until 1985 Jose Ramos-Horta became the representative of the Fretilin at the UN. Jose spoke to many different councils, committees and commissions about the human rights violations by the Indonesian military. He had a peace plan that would end all the violence in his country. During his time in exile, he was trained in human rights law in Australia. In February 1996 Jose was awarded the first UMPO prize, Unrepresented National and Peoples Organisation, for his â€Å"unswerving commitment to the rights of and freedoms of threatened people† later on it that year Jose Ramos-Horta and Bishop Carlos Belo were jointly awarded the Nobel Peace Prize. In 2006 Jose Ramos-Horta take a place in the government as caretaker prime minister after many hiccups with the previous government. Methods: The full-length sequence of EAV N gene was cloned by RT-PCR and ligated into pET32a vector for expression. The recombinant pET-N protein was expressed in E. coli and purified by Ni affinity chromatography. The purified N protein was used to immunize mice for preparing monoclonal antibody (mAb). The reactivity of mAb was evaluated by Western blot and immunofluorescence assay (IFA). The peptides were identified using the prepared mAb by indirect ELISA and Western blot. The homology analysis was performed using DNAMAN software. Results: Recombinant EAV N protein was successfully expressed in the procaryon expression system. An EAV N-reactive mAb was selected and designated as 1C11. Indirect ELISA results showed that overlapping domain of MBP-N10 and MBP-N11 was recognized by the mAb 1C11. Further, the indirect ELISA and Western blot showed that 101QRKVAP106 was the minimal linear epitope of the EAV N protein. EAV was à ¯Ã‚ ¬Ã‚ rst isolated from horses in Ohio in 1953. It is the prototype virus of the family Arteriviridae (genus Arterivirus, order Nidovirales) [4, 5]. EAV infection of horses has been reported in many countries including New Zealand, Australia, and South Africa [6-10]. EAV is a positive-sense, enveloped and single-stranded RNA molecule with a length of 12.7kb [11]. It contains two large open reading frames (ORFs, 1a and 1b) and seven smaller ORFs (2a, 2b, and 3 to 7). ORFs 1a and 1b encode two replicase polyproteins (pp1a and pp1ab), whereas the ORFs 2a, 2b, 5, 6, and 7 encode the known EAV structural proteins E, GS, GL, M, and N, respectively [12]. Moreover, ORFs 3 and 4 encode glycosylated membrane-associated proteins whose functional role is still under debate [13, 14]. All animal studies were approved by the Animal Ethics Committee of Harbin Veterinary Research Institute of the Chinese Academy of Agricultural Sciences (SYXK (H) 2006-032). Cell lines and virus SP2/0 myeloma and Rabbit kidney 13 (RK-13) cells were cultured and maintained in Dulbecco’s modified Eagle’s medium (DMEM; Invitrogen, Carlsbad, CA, USA) in a humidified 5% CO2 atmosphere at 37 °C. All culture media were supplemented with 10% heat-inactivated fetal bovine serum (GIBCO, Invitrogen) and antibiotics (0.1mg/ml streptomycin and 100 IU/ml penicillin).The Bucyrus strain of EAV (GenBank accession No. NC-002532.2, a highly cell culture-adapted strain provided by the key laboratory of Tropical and Subtropical Animal Viral Diseases in Yunnan province, China) was propagated in RK-13 cells and stored at -80à ¢Ã¢â‚¬Å¾Ã†â€™. Expression and characterization of recombinant EAV N protein The full-length sequence of EAV N gene was cloned by RT-PCR using the following primers: 5†²-CCCGGATCCATGGCGTCAAGACGATC-3†² (upstream) and 5†²-TTTGTCGACTTACGGCCCTGCTGGAGGCGCAAC-3†² (downstream). The primers contained BamH I and Sal I restriction sites (italicized). The purified and digested PCR product was ligated into an expression vector pET32a (Novagen, Germany). The pET-N recombinant plasmid was transformed into E. coli BL21 (DE3) and 1mM isopropyl-ÃŽ ²-D-1-thiogalactopyranoside (IPTG, Invitrogen, USA) was used for inducing expression of N protein. The recombinant proteins were obtained from the bacterial lysates. The insoluble inclusion bodies were washed and solubilized with phosphate buffered saline (PBS, pH 7.4). Then, the recombinant N protein fused with 6 His-tags was evaluated by SDS-PAGE and purified by Ni affinity chromatography according to manufacturer’s instruction (Invitrogen). Preparation and characterization of mAbs against N protein EAV N-reactive mAb was generated as previously described [17]. Briefly, 6-week-old female BALB/c mice were immunized with the purified recombinant N protein (100ÃŽ ¼g per mouse) mixed with an equal volume of Freund’s complete adjuvant (FCA, Sigma, USA). Two booster injections containing the same amount of purified N protein in an equal volume of Freund’s incomplete adjuvant (FICA) were given at 2-week intervals. The purified N protein without adjuvant was injected intraperitoneally as the final immunization. After three days of the final injection, the mice were euthanized and their splenocytes were fused with SP2/0 myeloma cells using polyethylene glycol (PEG4000, Sigma). The hybridoma cells were seeded into 96-well plates and selected in hypoxanthine-aminopterin-thymidine (HAT) selection medium (DMEM containing 20% fetal bovine serum, 100g/ml streptomycin, 100IU/ml penicillin, 100mM hypoxanthine, 16mM thymidine, and 400 mM aminopterin). After 5 days, the medium was re moved and replaced with hypoxanthine-thymidine (HT)-DMEM medium (DMEM containing 20% fetal bovine serum, 100g/ml streptomycin, 100IU/ml penicillin, 100mM hypoxanthine, and 16mM thymidine). After selection in HAT and HT medium, hybridoma supernatants were screened for evaluating reactivity and specificity of mAb by Western blot and immunofluorescence assay (IFA). Each MBP-fused polypeptide was induced by IPTG and screened by indirect ELISA. Briefly, MBP tags and purified N protein were used as negative and positive controls, respectively. Ninety-six-well microtiter plates were coated with expressed MBP-N fusion proteins at 4à ¢Ã¢â‚¬Å¾Ã†â€™ overnight and blocked with 5% skim milk for 1 h at 37à ¢Ã¢â‚¬Å¾Ã†â€™. For Western blot, the purified MBP-N recombinant proteins were electrophoresed on SDS-PAGE, and then transferred to a nitrocellulose membrane. Nonspecific antibody binding sites on the membrane were blocked with 5% skim milk in PBS overnight at 4à ¢Ã¢â‚¬Å¾Ã†â€™. The membrane was washed and incubated with mAbs for 1h at 37à ¢Ã¢â‚¬Å¾Ã†â€™. The membrane was incubated with HRP-conjugated goat anti-mouse IgG secondary antibody after five times washing with PBST. As shown in Fig.1b, c, mAb 1C11 reacted with recombinant N protein and total protein of EAV (Fig.1b, c). The reactivity of mAb was also assessed using RK-13 cells by IFA (Fig.1d). Identification of EAV N epitope To localize linear antigenic epitopes within the N protein, 11 16-amino acid long MBP fused peptides (MBP-N1 MBP-N11) were expressed and probed by mAb 1C11 by indirect ELISA. The results showed that MBP-N10 (91TVSWVPTKQIQRKVAP106) and MBP-N11 (95VPTKQIQRKVAPPAGP110) epitopes were recognized by the mAb 1C11 (OD450 > 1) (Fig. 2a). All the left fragments (MBP-N1-9) failed to react with the mAb. Because adjacent epitopes have 12 overlaps, we deduced that the linear epitope located in the overlapping domain of MBP-N10 and MBP-N11 (95VPTKQIQRKVAP106). Epitope mapping using mAbs has become a powerful tool to study protein structure and provides new tools to diagnose diseases and design vaccines [18]. Here, we defined one peptide epitope of EAV N protein in by using an EAV N-specific mAb. To our knowledge, epitope on the N protein of EAV has been published, but no previous studies about 101QRKVAP106 have been reported. Starick et al. [19] produced a mAb against the N protein to detect EAV. Weiland et al. [20] used the same method to produce a mAb against the N protein of EAV and to distinguish different virus isolates from semen and tissue samples after passaging through RK-13, Vero and fetal equine kidney cells. However, the minimal epitope of these mAb was not defined precisely. Similar to the work of Starick et al. and Weiland et al. [19, 20], a mAb named 1C11 against EAV N protein was prepared by using recombinant N protein expressed in E. coli and used for identifying B-cell epitopes on EAV N protein. mAb 1C11 reacted well with EAV by WB and IFA, thus this antibody may be a useful detection tool in EAV diagnosis. mAbs are useful and effective for mapping antigenic epitopes of viral proteins. In this study, for epitope mapping, 11 overlapping peptides from EAV N protein were expressed with MBP tags and identified by ELISA to screen linear epitopes. The ELISA results showed that the epitope located in the sharing region of MBP-N10 (91TVSWVPTKQIQRKVAP106) and MBP-N11 (95VPTKQIQRKVAPPAGP110). Then this region (95VPTKQIQRKVAP106) was expressed, and 7 peptides with deletions were obtained to identify the precise epitope. According to the results of ELISA and Western blot, 101QRKVAP106 was considered as the minimal linear epitope of EAV protein. This result is different from the previous studies [15, 21] which stated that the precise epitope of N protein located in amino acids 1-69. This may be due to the difference of the specificity and reactivity of the mAbs. Sometimes, a mAb can react with different locations of a viral protein. Sequence alignment showed that the identified epitope is very conservative among distinct regional EAV strains, but with a mutation of one amino acids on the ARVAC N protein epitope. This result suggests a slight regional difference emerged in this epitope. Therefore, it is possible to distinguish anti-Bucyrus EAV antibody from anti- ARVAC EAV antibody by using the epitope as antigen. This will be helpful in distinguishing the distinct regional EAV infection. This finding indicates that the N epitope of EAV identified in our study have a potential use in serological monitoring and differential diagnosis. In conclusion, one EAV N-specific mAb was developed and a minimal linear peptide epitope within the N protein was defined. The EAV N-specific mAb and the defined linear peptide epitope of EAV N protein may be useful for the development of specific diagnostic tools and design of vaccine.
